CutSharp Buffer是一种能让超过200种限制性内切酶的活性达到100%的缓冲液。使用CutSharp Buffer,可以免去内切酶酶切时选择buffer的麻烦,特别是在双酶切时会变得更加简单易行。此外,许多DNA修饰酶在CutSharp Buffer中也保留了100%酶活性,这样在酶切后进行DNA修饰酶处理就无需进行DNA纯化了。


各种内切酶和修饰酶在1×CutSharp buffer中的活性,请参考附录1和附录2。不同来源的限制性内切酶对于CutSharp Buffer的兼容性可能略有不同。1×CutSharp Buffer的组分为50mM Potassium acetate,20mM Tris-acetate,10mM Magnesium acetate,100μg/mL BSA,pH7.9@25℃,与NEB公司的CutSmart Buffer完全一致。
